The protocol of Ulcerative Colitis Drosophila Modeling using Dextran sodium sulfate (DSS)

Dextran sulfate sodium (DSS) is a polyanionic derivative of dextran produced by the esterification of Dextran with chlorosulphonic acid. Feeding Drosophila will destroy the connection between intestinal epidermal cells in the intestinal epithelium, causing intestinal damage, causing multiple pathways to respond to regulate the self-renewal and differentiation of stem cells, replenish damaged cells, and maintain intestinal homeostasis.

1 Model building

1.1 Animal Model

Drosophila, female, 5-10 days;

1.2 Material

DSS(Yeasen#60316ES,MW36000-50000)

1.3 Protocol

a) Feeding medium was prepared with 5% sucrose solution;
b) Control group: 5% sucrose medium (SC), Experimental group: 3% DSS (medium preparation);
c) Flies were cultured in an empty vial containing a piece of 2.5×3.75-cm chromatography paper (Fisher) wet with 5% sucrose solution as feeding medium;
d) Drosophila was cultured at 29°C for three days with a daily change of culture bottle;
e) After three days of culture, the midguts were dissected out and immunostained with GFP (green) and PH3 (red) antibodies and DRAQ5 (blue).

By using different labeling methods in immunostaining, the intestinal stem cells (ISC) and the proliferation of ISC precursor cells were quantified, indicating that DSS induced intestinal damage in the drosophila model.

4. Successful case for modeling with Yeasen DSS
